Objective: Sulfur mustard (SM), also termed mustard gas, is a potent vesicant that has been employed as a chemical weapon in various conflicts during the 20th century. It functions as a powerful alkylator and highly oxidative stress in both humans and animals More recently, mustard was used in the Iraq conflict against Iranian troops and civilians. At the present time there are more than 40.000 people suffering from pulmonary lesions special chronic obstructive pulmonary disease (COPD) due to mustard gas in Iran. SM increases the endogenous production of reactive oxygen species (ROS). The toxicity of oxidative stress reduces by antioxidant and there are adequate evidences that antioxidants have roles in cellular protection against oxidative stress. The present study was undertaken to investigate the expression of some antioxidant such as: HOs, NGAL, GST, and MT in bronchial epithelium in the human airway wall and compare with control group. mRNA expression of HO1 and HO2 also evaluated in both groups. We are going to propose a possible role that these molecules may play important role in cellular protection against oxidative stress due to mustard gas. Materials and Methods: 12 normal and 4 SM induced COPD patient bronchial biopsy were analysis by immunohistochemistry and 10 normal and 20 SM induced COPD bronchial biopsy used for mRNA expression of only HOs were performed by semiquantitative RT-PCR, real-time RT-PCR. Results: All SM exposed samples were immunonegative for HOs, NGAL, GST, MT antibodies in compare to unexposed patients, while Normal control samples expressed same level of HOs, expression level of HO1 and HO2 were upregulated about 1.35-4.27 folds respectively in the patients compared to normal samples which indicate SM induce GST expression. Conclusion: Our results revealed that all mentioned antioxidants expression were dramatically downregulated in compare with control group. Therefore its speculate that these antioxidants may plays an important role in cellular protection against oxidative stress due to mustard gas toxicity in airway of patients
